Peptide Reconstitution: Solvent Selection and Concentration Calculations

Reconstitution is where a lot of avoidable variability enters peptide research: the wrong solvent, an imprecise dilution, or the wrong storage temperature after mixing can all affect what you're actually working with. None of the steps are complicated, but they're worth getting right.

Choosing a Reconstitution Solvent

Most lyophilized peptides are reconstituted in bacteriostatic water, sterile water, or a dilute acetic acid solution, and the choice depends on the compound and how long the solution needs to remain stable.

Bacteriostatic Water vs Sterile Water

Bacteriostatic water contains a small amount of benzyl alcohol, which inhibits bacterial growth and extends how long a reconstituted solution can be stored under refrigeration. Sterile water contains no preservative and is generally used when a solution will be used quickly or when benzyl alcohol could interfere with a specific assay. For peptides sensitive to benzyl alcohol, sterile water is the safer default.

When a Co-Solvent Is Needed

Some peptides don't fully dissolve in plain water because of their charge or hydrophobicity. A dilute acetic acid solution (commonly 0.1% to 1%) is used for peptides that need a lower pH to stay in solution. Adding the acid solution slowly, rather than all at once, reduces the risk of localized precipitation.

Calculating Concentration

The basic relationship is straightforward: concentration in mg/mL equals the mass of peptide in the vial divided by the volume of solvent added. A 5 mg vial reconstituted with 2 mL of solvent gives a 2.5 mg/mL solution. The two mistakes that most often throw this off are misreading the vial's labeled mass and using an imprecise syringe for volume. A 1 mL syringe marked in 0.01 mL increments gives far more accurate dilutions than eyeballing a larger syringe.

Storage After Reconstitution

Once a peptide is in solution, it's no longer as stable as it was lyophilized. Reconstituted solutions should generally be kept refrigerated (2 to 8 degrees C) and used within the timeframe noted for that specific compound, often days to a few weeks, not months. Repeated freeze-thaw cycles degrade most peptides faster than steady refrigeration, so aliquoting into single-use portions before freezing is preferable to refreezing a partially used vial.

A Few Practical Checks

  • Add solvent slowly, down the side of the vial, rather than directly onto the lyophilized powder
  • Let the vial sit briefly rather than shaking vigorously, which can stress some peptide structures
  • Label reconstituted vials with the date and calculated concentration, not just the compound name
  • Confirm the specific storage window for that peptide rather than assuming a standard timeframe applies
